External Recording Solution for Voltage-Gated Ca2+Channels

Reagent Final concentration
Methanesulfonic acid (CH3SO3H)   140 mm
Tetraethylammonium hydroxide (TEA-OH)   145 mm
HEPES    10 mm
d-glucose    15 mm
CaCl2·2H2O    10 mm
Tetrodotoxin (TTX)  0.0003 mm
Prepare solution containing all but TTX in deionized water. Adjust pH to 7.4 with TEA-OH. (Osmolality should range between 310 and 325 mmol/kg.) Filter using a 0.2-µm-pore membrane filter. Store up to 3 mo at room temperature (20°C–22°C) or up to 6 mo at 4°C.
Add TTX before the start of recordings. Be aware that TTX is susceptible to breakdown under basic (high-pH) conditions.
