Internal Recording Solution for Voltage-Gated Ca2+Channels

Reagent Final concentration
HCl  20 mm
N-methyl-d-glucamine (NMG) 120 mm
Tetraethylammonium hydroxide (TEA-OH)  20 mm
EGTA  11 mm
HEPES  10 mm
Sucrose  10 mm
CaCl2  1 mm
Tris-creatine phosphate  14 mm
Mg-ATP  4 mm
Na2-GTP  0.3 mm
Prepare in deionized water. Adjust pH to ~8.5 with methanesulfonic acid (CH3SO3H) before adding Tris-creatine phosphate, Mg-ATP, and Na2-GTP. Adjust final pH to 7.2 with methanesulfonic acid (CH3SO3H). (Osmolality should range between 290 and 305 mmol/kg.) Filter internal recording solution using a 0.2-µm-pore membrane filter. Divide into 5-mL aliquots and store up to 6 mo at −20°C.
