X-Gal Plates

| Yeast nitrogen base without amino acids/ammonium sulfate | 1.7 g |
| Ammonium sulfate | 5.0 g |
| Dextrose | 20.0 g |
| Agar | 20.0 g |
| Dropout powder | 0.8 g |
| NaOH | one pellet |
| Combine ingredients, add water to 898 mL, and autoclave. Cool to 45°C–50°C. Add 100 mL of 0.7 m potassium phosphate (pH 7.0). Add 2 mL of X-gal solution (20 mg/mL in 100% N,N-dimethylformamide). Pour the plates while the solution is still warm. | |
