5-Fluoroorotic Acid (5-FOA) Plates

0.67% yeast nitrogen base

0.2% SC–Ura drop-out mix

2% glucose

50 µg/mL uracil

0.1% 5-FOA

Combine the ingredients above, and add distilled water to give a total volume of 500 mL. Filter-sterilize using a 0.2-µm filter. Dissolve 10 g of bacto-agar in 500 mL of distilled water, autoclave, and cool to ∼80°C. Mix the two solutions, and pour the plates while the solution is still warm. Store the plates for up to 3 mo at 4°C.
