SPO Agar

• 1. Combine the following reagents in deionized H₂O.

  Reagent	Final concentration
  Agar	2%
  Potassium acetate	2%

• 2. Sterilize by autoclaving for 20 min at 15 psi.

• 3. Add trace nutrients appropriate for the yeast background of interest.

• 4. Pour 20–25 mL of agar medium per 90 × 15-mm Petri dish.
