YPD Agar

• 1. Combine the following reagents in deionized H₂O.

  Reagent	Final concentration
  Agar	2%
  Peptone (BD 211677)	2%
  Yeast extract (BD 212750)	1%

• 2. Prepare a solution of 20% glucose in deionized H₂O.

• 3. Sterilize both solutions by autoclaving for 20 min at 15 psi.

• 3. Add the sterile 20% glucose solution to the medium from Step 1 to a final concentration of 2% glucose.

• 4. Pour 20–25 mL of agar medium per 90 × 15-mm Petri dish.
