Bottom and Top Layer Media for Colony-Forming Assay
| Cell culture medium (2×) | 45 mL |
| Fetal bovine serum (FBS) | 9 mL |
| Glutamine (200 mm) | 1 mL |
| Prepare fresh 2× cell culture medium appropriate for the cell line of interest using a powdered medium (e.g., Life Technologies 3180089) and filter-sterilize. Then add FBS and glutamine. Prepare sufficient volume for the samples to be assayed. One-half of this recipe is to be used for the bottom layer and the other half for the top. | |
