CsF/CsCl Recording Solution

Reagent Amount Final concentration
CsF 1.329 g  35 mm
CsCl 4.209 g 100 mm
EGTA 0.951 g  10 mm
HEPES 0.596 g  10 mm
In a 250-mL volumetric flask, combine the above reagents. Add ∼200 mL of H2O, stirring constantly. Adjust the pH to 7.3 using CsOH solution. Measure the osmolarity of the CsF/CsCl recording solution using an osmometer. Adjust to ∼300 mOsm by adding H2O. Verify the pH after adjustment of osmolarity, adding CsOH or HCl if necessary. Store the solution at 4°C for up to several months. (This recording solution is optimized for simple voltage-clamp recordings. Different recording solutions are appropriate for different experimental goals. QX-314 [2 mm] may be added to block sodium channels and improve clamp.)
