Artificial Cerebrospinal Fluid (ACSF) (10×)

Reagent Amount to add Final concentration in 10× ACSF Final concentration in 1× ACSF
NaCl  73.05 g  1.25 m  125 mm
NaHCO3 21.843 g   260 mm  26 mm
NaH2PO3  1.725 g  12.5 mm 1.25 mm
KCl  1.864  25 mm  2.5 mm
  • In a 1-L volumetric flask, combine the above reagents. Shake to prevent poorly soluble monobasic crystals from forming. Add MgCl2 from a concentrated stock solution to a final concentration of 10 mm (in 10× ACSF). (For example, add 5 mL of a 2 m MgCl2 stock.) Add H2O to the 1-L line and stir until all reagents are dissolved. Store 10× ACSF in a 1-L flask at 4°C for up to 2 wk. Use this 10× stock solution daily to prepare Artificial Cerebrospinal Fluid (ACSF) (1×). Do not use if crystals form. (10× ACSF can also be prepared without MgCl2 for experiments requiring different calcium concentrations.)
