  • 1. Add each chemical in order and mix carefully. Note that 60% syrup of sodium lactate has a large osmotic effect and must be measured very accurately.

  • 2. Dissolve CaCl2 in 5 mL of H2O separately and add it to the solution.

  • 3. Before adding BSA, apply gas for 5 min with 5% CO2, 5% O2 balanced with N2, using a 1-mL pipette submerged in the medium or place in the incubator.

  • 4. Add BSA after gassing, and incubate to dissolve.

  • 5. Filter through a 0.22-µm filter.

  • 6. Measure osmolarity; it should be 290% ± 5% Osm.

  • 7. Gas before refrigerating.

  • 8. Store no longer than 2 wk at 4°C. Can be kept frozen for 6 mo or more depending on the temperature.

  • 9. Always gas medium with CO2 after every use. The medium should be salmon pink at the time of use.
