Synthetic Dextrose (SD) Plates (with or without Cytosine)

Mix yeast nitrogen base with ammonium sulfate and without amino acids (6.7 g/L), dextrose (20 g/L), and Bacto agar (20 g/L) in 1 L of (deionized or distilled) H2O. (Other nutritional supplements may be required depending on strain genotypes; if necessary, dropout mixtures may be made as described [Rose et al. 1990] or obtained from commercial sources such as ClonTech.) Autoclave. If necessary, add filter-sterilized cytosine to a final concentration of 25 µm (Hartzog et al. 2005) after autoclaving. After cooling to 55°C–60°C, pour into Petri dishes.
