Synthetic Dextrose Glutamate (SDE) Plates with Drugs
Mix yeast nitrogen base without ammonium sulfate and without amino acids (1.7 g/L), l-glutamate (monosodium salt; 1 g/L), dextrose (20 g/L), and Bacto agar (20 g/L) in 1 L of (distilled or deionized) H2O, and autoclave. After autoclaving, add one of the following filter-sterilized drugs, as required, to the appropriate final concentration: G418 (200 µg/mL), hygromycin B (300 µg/mL), or clonNAT (100 µg/mL). After cooling to 55°C–60°C, pour medium into Petri dishes. If using glutamic acid instead of the suggested monosodium glutamate, adjust medium pH to 5.5 before autoclaving.
