YPD Liquid Medium and Plates with Drugs
For YPD liquid medium, mix yeast extract (10 g/L), Bacto peptone (20 g/L), and dextrose (20 g/L) in 1 L of (deionized or distilled) H2O. Adjust the pH to 5.5 with 1 m HCl, and autoclave.
For YPD plates with drugs, prepare liquid medium as above, but add Bacto Agar (20 g/L) before autoclaving. After autoclaving, add one of the following filter-sterilized drugs, as required, to the appropriate final concentration: clonNAT (100 µg/mL), G418 (200 µg/mL), hygromycin B (300 µg/mL), or phleomycin (7.5 µg/mL). After cooling to 55°C–60°C, pour liquid into Petri dishes.
