YPD + 5-Fluorocytosine Medium

1. Autoclave YPD (1% yeast extract, 2% Bacto peptone, 2% dextrose, and 2% Bacto agar).

2. After autoclaving, add filter-sterilized 5-fluorocytosine to a final concentration of 1 mM, mix thoroughly, and pour the medium into Petri dishes.

5-Fluorocytosine solutions should be made up fresh to limit spontaneous deamination to 5-fluorouracil, which reduces the effectiveness of the counter selection for resistance to 5-fluorocytosine. 5-Fluorocytosine-containing media are best used fresh but, if necessary, can be stored for several months at 4°C in the dark.
