N2B27 Medium with LIF and 2i Inhibitors

Reagent Amount Final concentration
N2B27 medium 50 mL
LIF (1 × 107 units/mL) (Millipore ESG 1107)  5 µL 1000 units/mL
CHIR99021 (10 mm)  15 µL 3 µm
PD0325901 (10 mm)  3 µL 1 µm
Once 2i inhibitors and LIF are added, the medium can be stored for no more than 1 wk.
