Synthetically Defined (SD) Double-Selective Drop-Out Medium

Reagent Quantity (for 1 L)
Yeast nitrogen base (without amino acids and nitrogen substrate)  1.7 g
Ammonium sulfate   5 g
Dextrose   20 g
Complete supplement mixture (CSM) of amino acids and nucleotides Varies
Agar (for solid plates)   20 g
Dissolve the yeast nitrogen base, ammonium sulfate, and dextrose in water. In amounts specified by the supplier, dissolve the CSM (minus those amino acids or nucleotides used for selection) in the solution. Add the agar (if preparing plates). Bring the final volume to 1 L with water. Autoclave for 20 min at 121°C and 0.5 bar. Store for several weeks at room temperature.
