Synthetic Minimal Solid Medium

  • 1. In a 0.5-L bottle, combine 200 mL of H2O and 10 g of agar. In a 250-mL bottle, combine 3.4 g of yeast nitrogen base (YNB) without amino acids (Sigma-Aldrich Y0626) and 200 mL of H2O. Autoclave each solution for 30 min at 15 psi and then combine.

    • It is critical to autoclave agar and YNB separately, as the low pH of YNB will hydrolyze the agar and yield semisolid plates.

  • 2. Add 25 mL of 40% (w/v) glucose (separately sterilized by autoclaving).

  • 3. Pour plates as required for use.
