Synthetic Minimal Solid Medium

1. In a 0.5-L bottle, combine 200 mL of H2O and 10 g of agar. In a 250-mL bottle, combine 3.4 g of yeast nitrogen base (YNB) without amino acids (Sigma-Aldrich Y0626) and 200 mL of H2O. Autoclave each solution for 30 min at 15 psi and then combine.

It is critical to autoclave agar and YNB separately, as the low pH of YNB will hydrolyze the agar and yield semisolid plates.

2. Add 25 mL of 40% (w/v) glucose (separately sterilized by autoclaving).

3. Pour plates as required for use.
