β-Gal Reaction Buffer

20 mm K3Fe(CN)6

20 mm K4Fe(CN)6·3H2O

2 mm MgCl2

1 mg/mL X-gal (5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside)

0.1 m phosphate-buffered saline with 0.05% Tween (PBT)

Prepare immediately before use. X-gal will create a blue reaction product that nicely contrasts with red in situ hybridization chromogens. Alternatively, Magenta-gal (5-Bromo-6-chloro-3-indolyl β-D-galactopyranoside) or Red-gal (6-Chloro-3-indolyl-β-D-galactopyranoside) can be used to contrast with blue/purple chromogens.
