Nuclear Preparation Buffer (NPB) Solutions Containing Bovine Serum Albumin (BSA)

  • 1. To prepare a solution of 10% BSA in NPB, dissolve 2 g of BSA in 15 mL of 1× NPB. Adjust the pH to 7.6 with KOH, and bring the volume to 20 mL with 1× NPB.

  • 2. To prepare a solution of 3% BSA in NPB, combine 10.5 mL of 1× NPB with 4.5 mL of 10% BSA in NPB for a total of 15 mL.

  • 3. To prepare a solution of 0.3% BSA in NPB, combine 9 mL of 1× NPB with 1 mL of 3% BSA in NPB for a total of 10 mL.

  • 4. To prepare a solution of 0.3% BSA in NPB plus 30% glycerol, combine 3.5 mL of 0.3% BSA in NPB with 1.9 g of glycerol, and bring the total volume to 5 mL with 0.3% BSA in NPB.

    • Prepare all NPB/BSA solutions fresh, and use the same day. Store on ice until use.
