| These two polyamines are highly hygroscopic. Therefore, avoid weighing these compounds. Instead, order preweighed small bottles
(e.g., 1 g), dissolve each polyamine in the appropriate amount of sterile H2O, mix the two polyamine solutions into one tube, sterilize through a 0.22-µm filter, aliquot into 500 µL portions, and store
at −20°C, where it can be kept for >1 yr. Working concentrations for 1× polyamines are 30 µm spermine and 70 µm spermidine. For YAC equilibration and YAC microinjection buffers, use the 1000× polyamine mix to obtain the 1× working solution.
