Modified Leibovitz's L-15 Medium

1. Add 13.8 g of L-15 powdered medium (Leibovitz's L-15 medium, with glutamine; Sigma-Aldrich L4386) to 1300 mL of dH2O. Mix well.

2. Adjust pH to 7.6 with 1 n NaOH or 1 n HCl.

3. Add 149 mL of fetal bovine serum (FBS, certified, heat-inactivated; Gibco 10082147).

4. Adjust the final volume to 1.493 L.

5. Filter-sterilize with a 0.2-µm surfactant-free cellulose acetate membrane filter unit.

6. Freeze the medium or maintain it at 4°C for up to 4 wk.
