Steinberg's Solution (2×)

Reagent Quantity needed for 50 mL of stock solution Final concentration of stock solution Volume of stock solution needed for 2× Steinberg's solution
NaCl  8.5 g   17% 2 mL
KCl 0.25 g  0.5% 1 mL
Ca(NO3)2•4H2O  0.4 g  0.8% 1 mL
MgSO4•7H2O 1.03 g 2.05% 1 mL
Tris  2.8 g  100× 1 mL
Prepare 50 mL each of NaCl, KCl, Ca(NO3)2, MgSO4, and Tris stock solutions in dH2O, and filter to sterilize. Store stock solutions at room temperature. Before each use, freshly prepare 2× Steinberg's solution (50 mL) by combining the volumes of stock solutions listed, adjust the pH to 7.5 with sterile 1 n HCl, and then bring to a final volume to 50 mL with sterile dH2O.
