1.25× Stock Solution II

137.5 mm KCl (diluted from 1 m solution, e.g., Sigma-Aldrich 60142)

62.5 mm Tris-HCl (pH 7.4) (diluted from 1 m solution, e.g., Fisher BioReagents BP1757-500)

6.25 mm MgCl2 (diluted from 1 m solution, e.g., Sigma-Aldrich M1028-100ML)

0.125 mm EDTA (diluted from 500 mm solution, e.g., Invitrogen 15575020)

2.75 m sucrose (e.g., Sigma-Aldrich S0389-1KG)

Prepare with RNase/DNase-free H2O. It takes time to dissolve sucrose at this concentration. As needed, heat the solution and vortex vigorously. Filter-sterilize the solution with a 0.2 µm filter. Store for up to 6 mo at 4°C. If the sucrose becomes crystalized during storage, heat the solution and dissolve the crystals completely before use.
