Albumin-Gelatin Embedding Medium

  • 1. Carefully dissolve 24.2 g albumin (Sigma-Aldrich A5253) in 66 mL of distilled water. Use a magnetic stirrer to slowly dissolve the albumin in the water, and add the albumin gradually (over the course of 1 h) to avoid the formation of clumps or foam. Try to dissolve any clumps using a spoon. After albumin has been dissolved, stir gently for another 30 min and discard any foam that forms.

  • 2. Warm 25 mL of distilled water to 55°C and add 5.7 g of gelatin (Sigma-Aldrich G2500). Place the solution on the magnetic stirrer, keep it warm, and gently mix until gelatin has been completely dissolved.

  • 3. Add the dissolved gelatin to the albumin solution while slowly stirring on the magnetic stirrer for ∼5 min.

  • 4. Divide into 15-mL aliquots in 50-mL Falcon tubes and freeze at −20°C until used.
