Albumin-Gelatin Embedding Medium

1. Carefully dissolve 24.2 g albumin (Sigma-Aldrich A5253) in 66 mL of distilled water. Use a magnetic stirrer to slowly dissolve the albumin in the water, and add the albumin gradually (over the course of 1 h) to avoid the formation of clumps or foam. Try to dissolve any clumps using a spoon. After albumin has been dissolved, stir gently for another 30 min and discard any foam that forms.

2. Warm 25 mL of distilled water to 55°C and add 5.7 g of gelatin (Sigma-Aldrich G2500). Place the solution on the magnetic stirrer, keep it warm, and gently mix until gelatin has been completely dissolved.

3. Add the dissolved gelatin to the albumin solution while slowly stirring on the magnetic stirrer for ∼5 min.

4. Divide into 15-mL aliquots in 50-mL Falcon tubes and freeze at −20°C until used.
