Sperm Storage Buffer for Xenopus (SSB)

500 µL 2× NPB

300 µL glycerol (e.g., Fisher G33500)

170 µL sterile Nanopure H₂O

30 µL 10% bovine serum albumin (BSA; e.g., Sigma-Aldrich A3294; 10% w/v with pH adjusted to 7.6 with KOH, aliquoted, and stored at −20°C)

Freshly prepare 1 mL and store on ice until use. Alternatively, for a larger volume, aliquot and store for a year at −20°C.
