Antibody Dilution Buffer for Whole-Mount Immunohistochemistry

  • 1. In a 15-mL Falcon tube, combine 200 mg of bovine serum albumin (final concentration 2%, w/v), 9.56 mL of Dulbecco’s phosphate-buffered saline (DPBS; Wisent), 200 µL of 10% (v/v) normal sheep serum (Jackson ImmunoResearch Inc.), and 40 µL of Triton X-100.

  • 2. Vortex this mixture well. Makes 10 mL.

    • For optimal results, freshly prepare; however, aliquots can be made and stored long term (up to 1 yr) at20°C.
