Mowiol Solution

  • 1. Stir 12 g of glycerol (e.g., AnalaR Normapur 24388.295) and add 4.8 g Mowiol (e.g., Sigma-Aldrich 81381) while stirring. Add 12 mL Milli-Q H2O and let stir for 2 h at room temperature.

  • 2. Heat 24 mL of 0.2 m Trizma base (2.42 g Trizma base in 100 mL Milli-Q water; e.g., Sigma-Aldrich T1503), pH 8.5, to 50°C.

  • 3. Heat the Mowiol solution to 50°C while stirring in a water bath and add the preheated Trizma base. Continue stirring for 10 min at 50°C.

  • 4. Divide the solution in 50-mL Falcon tubes and centrifuge for 25 min at 3000g at room temperature.

  • 5. Pour supernatant in a new Falcon tube, discard the pellet, and add 0.1% DABCO (e.g., Sigma-Aldrich D27802) to improve fluorescence lifetime.

  • 6. Stir lightly and prepare aliquots of 1 mL. Store the aliquots at −20°C for long-term storage or up to 2 wk at room temperature.
