Tricaine Stock (0.3%, w/v)
Prepare 1 m Tris by mixing 60.57 g of Trizma base (e.g., Sigma-Aldrich T1503) in 500 mL of Milli-Q H2O. Adjust to pH 9 using HCl, 2 n.
Add 14.7 mL of 1 m Tris to 685.3 mL of distilled H2O. Dissolve 2.1 g of Tricaine (MS-222, e.g., Sigma-Aldrich A5040) while stirring and adjust pH to 7 ± 0.02 using HCl 2 n).
Store up to 6 wk in a dark bottle at 4°C.
