Tris-NaCl Blocking Buffer (TNB)
Mix 5 g of TSA Blocking Reagent (PerkinElmer FP1012) in 1 L of Tris-buffered saline with 0.1% Triton. Add the Blocking Reagent slowly to the buffer while stirring. Heat to 55°C with continuous stirring to completely dissolve the Blocking Reagent (30–60 min). Freshly prepare or store up to 2 yr at −20°C, and thaw the needed volume just prior before use.
