Papain Solution
Add 250 µL of papain (from papaya latex; Sigma-Aldrich P3125), 100 µL DNase I solution (10 mg/mL), and 200 µL l-cysteine solution to 5 mL of DMEM/F12. Sterilize through a Millex 0.22-µm-pore-size filter (Fisher Scientific SLGP033RS).
Freshly prepare before use (important!). Papain degrades proteins that make up tight junctions between cells.
