LB Agar, Miller

1. Add 20 g of LB agar powder (BD 244520) to 500 mL of H2O in a 1-L bottle containing a magnetic stir bar.

2. Stir well to mix and subsequently autoclave using standard conditions.

3. (Optional) Once cooled to 50°C, add 500 µL of a 100 mg/mL ampicillin stock solution to give a final concentration of 100 µg/mL ampicillin and stir gently to mix well.

4. Once cooled to 50°C, pour ∼20–25 mL of medium into 90-mm round Petri dishes. Let the plates solidify, turn them lid-side down, and let them dry overnight at room temperature before use.

5. Store the plates for up to 1 mo at 4°C.
