TSA Chloramphenicol Egg Yolk Plates

  • 1. Crack an egg open and transfer the egg yolk into a 50-mL Falcon tube.

  • 2. Add an equal volume of sterile phosphate-buffered saline (PBS), pH 7.4, and vortex vigorously to mix.

    • Store the egg yolk suspension for approximately up to 1 wk at 4°C.

  • 3. Autoclave Tryptic Soy Agar (TSA) for 15 min at 121°C and 15 psi.

  • 4. After the agar has cooled down to 50°C, add 2 mL of the egg yolk–PBS mixture per 100 mL of autoclaved agar, and add chloramphenicol stock solution (10 mg/mL) to a final concentration of 10 µg/mL.

  • 5. Stir well to combine and cast 25 mL per 9-cm Petri dish. Allow to solidify and dry overnight at room temperature.

  • 6. Store the plates for up to 1 wk at 4°C.
