FISH Hybridization Buffer for Nothobranchius

Reagent stock Volume required for 50 mL Final concentration
Formamide, 100%
 (Sigma-Aldrich 221198-1L) 		25 mL 50%
Sodium chloride-sodium citrate buffer
 (SSC), 20× 		12.5 mL 5×
Tween-20 (VWR 97062-332),
 10% (v/v) stock 		250 µL 0.05%
tRNA, 50 mg/mL (Sigma-Aldrich
 R7876) 		500 µL 500 µg/mL
Heparin (Sigma-Aldrich H3393),
 50 mg/mL 		50 µL 50 µg/mL
H2O Adjust to 50 mL

  • Adjust to pH 6.0 with 1 m citric acid. Sterilization is not necessary. Store for up to a year at −20°C.
