Molten LB Top Agar

1. Prepare molten LB agar and LB (Luria-Bertani) liquid medium.

2. Cool LB agar and LB broth to 60°C.

3. Add two volumes of LB broth to one volume LB agar, and swirl to mix.

This reduces the concentration of agar from 1.5% to 0.5%.

4. Cool to 50°C before use.
Molten LB top agar may be kept for up to 1 wk at 60°C and must be cooled down to 50°C before use. Aliquots of molten LB top agar may be stored in glass bottles and kept for up to 6 mo at 4°C. When molten LB top agar is needed, melt in a microwave, and then cool to 50°C before use.
