YTCG Liquid Medium
Mix 16 g of tryptone, 10 g of yeast extract, and 5 g of NaCl. Add H2O and mix to dissolve all reagents. Adjust pH to 7.0 with NaOH. Adjust volume to 950 mL with deionized H2O. Sterilize by autoclaving for 20 min at 15 psi (1.05 kg/cm2) on liquid cycle. Allow medium to cool down. Right before use, add 1 mL of a sterile carbenicillin stock solution, for a final carbenicillin concentration of 100 µg/mL, and 50 mL of filter-sterilized 20% (w/v) glucose, for a final glucose concentration of 1% (w/v). YTCG liquid medium can be stored for 2 wk at 4°C.
