Egg-Collection Apple Juice Agar Plates

Reagent Quantity
Agar 30 g
Apple juice (100%) 333 mL
Double-distilled H2O 1000 mL
Methyl-4-hydroxybenzoate 2 g
Sucrose 30 g
  • 1. In a flask, microwave 30 g of agar and 1 L of double-distilled H2O until the agar is completely dissolved. Remove the solution from the microwave and cool to 60°C.

  • 2. Mix sucrose, apple juice, and methyl-4-hydroxybenzoate in another flask, and bring it to a boil in a microwave oven.

  • 3. Carefully combine the solutions, stir well, and pour 10 mL into 60-mm polystyrene Petri dishes. Let the agar completely solidify, and store at 4°C in sealed bags to prevent desiccation. Plates will stay fresh for 2 mo stored at 4°C.
