SB (Super Broth) Medium

Mix 20 g of 3-(N-morpholino)propanesulfonic acid (MOPS; MilliporeSigma M1254), 60 g of Bacto Tryptone (Thermo Fisher Scientific 211705), and 40 g of Bacto Yeast Extract (Thermo Fisher Scientific 212750). Add highly pure water (e.g., that produced by Hydro PicoPure UV Plus) to a final volume of 1.9 L. Bring to pH 7.0 with 1 m sodium hydroxide (NaOH; Thermo Fisher Scientific A4782901). Bring volume to 2 L with highly pure water. Sterilize by autoclaving for 15 min at 121°C and 15 psi in two 1-L or four 500-mL glass bottles. Store for up to 2 yr at room temperature if unopened.
