2× YT Plates
Add 7.5 g of Bacto Agar (BD 214010) to 500 mL of 2× YT medium. Autoclave for 15 min at 121°C and 15 psi. Pour into 10-cm sterile Petri dishes (20–25 mL each), remove bubbles by flaming the plates with a Bunsen burner, and allow to solidify at room temperature. Store for up to 3 mo at 4°C.
