Solution II for agarose plugs

~1 M Sorbitol (Merck, autoclaved)

20 mM EDTA (pH 8.0) (autoclaved)

2% SeaPlaque GTG agarose (FMC 50112), 0.2 g

14 mM β-mercaptoethanol (Merck)

sterile H₂O

Prepare fresh. Melt the Sorbitol plus EDTA and agarose in the microwave. Equilibrate in a water bath down to 37-40°C. Add the β-mercaptoethanol and keep the solution at 37-40°C until use.
