Trolox solution
MATERIALS
Reagents
Trolox (6-hydroxy-2,5,7,8-tetramethylchromane-2-carboxylic acid; Sigma-Aldrich)
M5 buffer is used in FIONA myosin motility assays.
Equipment
0.22-μm syringe filter (e.g., Millex GP; Millipore)
15-mL tube
METHOD

1. Transfer 5 mg of Trolox to a 15-mL tube.

2. Add 10 mL of the desired buffer (e.g., M5 for myosin motility experiments) to the tube and mix completely to saturate the buffer with Trolox (which is moderately soluble in H2O).

3. Filter the mixture through a 0.22-μm syringe filter.

4. Adjust the pH of the buffer to 7.5 with NaOH.
