Antibiotics (appropriate for selection of T-DNA)
Indolebutyric acid
1X MS salts (GIBCO)
2-[N-Morpholino]ethanesulfonic acid (MES)
Phytagel
Sucrose
For 1 liter of medium, dissolve 1X MS salts (GIBCO), 10 g of sucrose and 0.5 g of MES in water. Adjust the pH to 5.7 with 1 N KOH. Add 1.1 g of Phytagel. Autoclave the solution. Add 2 ml of 500X Vitamix, 2 mg of indolebutyric acid, 500 mg of Timentin and appropriate antibiotics for selection of the T-DNA of interest.
Use elevated levels of antibiotic for good selection of transgenic roots.
