Embryo staining solution

MgCl₂ (2 mM)

NP-40 (0.02%)

Phosphate buffer (0.1 M, pH 7.3), prepared using phosphate buffer tablets (e.g., BDH)

Potassium ferricyanide (5 mM)

Potassium ferrocyanide (5 mM)

Sodium deoxycholate (0.01%)

Dilute the 25 mg/ml X-gal stock into the solution above to give a final concentration of 1 mg/ml X-gal. After use, the stain can be filtered and reused; it is good for months. Store at 4°C protected from light.
