Sucrose density gradient solutions

PE buffer (50X)

Sucrose (Sigma S7903)

Triton X-100 (Sigma-Aldrich T9284), 10% (w/v) in H₂O

• 1. Prepare 150 mL of 1X PE buffer containing 0.1% (w/v) Triton X-100.

• 2. Prepare the sucrose solutions:

  • To prepare a 20% (w/w) solution:

  • i. Zero a container on a balance.

  • ii. Add 10 g sucrose to the container.

  • iii. Add the PE/Triton solution slowly until the total mass equals 50 g.

  • iv. Stir to mix.

  • To prepare a 62.5% (w/w) solution:

  • v. Zero a container on a balance.

  • vi. Add 31.25 g sucrose to the container.

  • vii. Add the PE/Triton solution slowly until the total mass equals 50 g.

  • viii. Stir to mix.
