AP reaction buffer for Parhyale

MgCl₂	5 mM
NaCl	100 mM
Tris (pH 9.5)	100 mM
Triton X-100	0.1%

Prepare just before use; buffer that has been sitting for a few hours at room temperature will not work as well. This buffer with a pH of 9.5 is used for BCIP/NBT reactions. For Fast Red and Vector Red reactions, a pH of 8.2 is optimal. This can be made by mixing 7 volumes of 1 M Tris (pH 9.5) with 13 volumes of 1 M Tris (pH 7.5) for a final concentration of 100 mM Tris (pH 8.2).
