Elongation medium

Recipe

Reagent	Amount to add (for 1 L)	Final concentration
Murashige and Skoog (MS) basal salt mixture with vitamins (Phytotechnology Laboratories)	4.3 g	0.43%
Sucrose	30 g	3%
Gibberellic acid (GA₃)	2 mg	0.2%
Agar	7.5 g	0.75%

Adjust pH of medium to 5.8 using 1 M NaOH.
Dissolve GA₃ in a few drops of 1 M NaOH before adding H₂O to make a working solution of 1 mg/mL.
Autoclave and cool to 50°C.
Add filter-sterilized (0.2-μm) antibiotics: 300 mg/L carbenicillin (to kill Agrobacterium LBA4404) 100 mg/L kanamycin (to select for positive transformants)
Store at 4°C. Medium is good for 1 mo.
