Rooting medium (pH 5.8)

Recipe

Reagent	Amount to add (for 1 L)	Final concentration
Murashige and Skoog (MS) basal salt mixture with vitamins (Phytotechnology Laboratories)	2.15 g	0.215%
Sucrose	30 g	3%
Agar	7.5 g	0.75%

Adjust pH to 5.8 using 1 M NaOH. Autoclave and cool to 50°C.
Add filter-sterilized (0.2-μm) antibiotics: 300 mg/L carbenicillin (to kill Agrobacterium LBA4404) 100 mg/L kanamycin (to select for positive transformants)
Pour ~40 mL per box into magenta boxes and store at 4°C. Medium is good for 2 mo.
