A human cell (U2OS) expressing a gene that transcribes an mRNA with 24 MS2 repeats in its 3′-UTR was cotransfected with the CFP-Lac repressor, YFP-MS2

The gene was transcriptionally induced (Tet-On doxycycline) for 30 min. (See Shav-Tal Y, Darzacq X, Shenoy SM, Fusco D, Janicki SM, Spector DL, Singer RH. 2004. Dynamics of single mRNPs in nuclei of living cells. Science 304: 1797-1800.) The movie shows the cell nucleus, in which the gene (DNA) is labeled in red (by CFP-Lac repressor binding) and the mRNAs transcribed from the gene are labeled in green (YFP-MS2).
