A human cell (U2OS) expressing a gene that transcribes an mRNA with 24 MS2 repeats in its 3′-UTR was cotransfected with the CFP-Lac repressor, YFP-MS2 (second example)

Movie 37_2.
The gene was transcriptionally induced (Tet-On and doxycycline) for 30 min. (See Shav-Tal Y, Darzacq X, Shenoy SM, Fusco D, Janicki SM, Spector DL, Singer RH. 2004. Dynamics of single mRNPs in nuclei of living cells. Science 304: 1797-1800.) The movie shows the cell nucleus and the YFP-MS2-tagged mRNPs. A transcription site is detected in the center of the nucleus, and the release of mRNPs is observed.Supplemental movie from Live Cell Imaging, 2nd ed., Cold Spring Harbor Laboratory Press.
