Adjust the pH to 7.4.
Add Tween 20 (1%, v/v).
If the buffer is to be used for immunostaining of Ca2+-binding structures, omit the EGTA and Mg2+ (i.e., just use Tris-buffered saline containing 1% Tween 20).
