Resolving gel mixture

9.4 mL H₂O

2.4 mL acrylamide stock (40% T, 2.67% C)

4 mL of 1.5 M tricine-NaOH (pH 8)

0.16 mL of 10% ammonium persulfate

16 μL TEMED

Mix the first four ingredients. Add the TEMED just before pouring the gel. This makes enough gel mixture to pour two minigels, 1.5-mm thick.

The final mixture is 6% acrylamide, 375 mM tricine-NaOH (pH 8).
